The Morning Show star was nominated for Outstanding Lead Actress but lost to Anna Sawai.
Despite the loss, the actress told the media she already felt like she had won.
However, it was her comment about her Friends character Rachel Green that caught fans' attention.
Aniston was rushing down the red carpet at the award ceremony but stopped to answer a question from Access Hollywood.
The presenter asked, "Just tell us, where would Rachel Green be right now 30 years later?"

Green responded: "She's right here" and pointed at herself.
"We're all the same," the actress added when talking about her iconic character.
Fans of the show were touched by how much the character still means to Aniston, despite the actress working on many other projects since the TV show aired.
Fans of the actress couldn't help but comment on how humble she was.
"She seems like a genuinely lovely person," one said.
One wrote: "Love her here & Now & on friends."
"It's so lovely to see a beautiful genuine smile on her face and a bounce in her step," another said.
"I love her! She's still so down to earth," another wrote.
Her appearance at this year's Emmys comes after Aniston honoured her late friend Matthew Perry.
In an interview with Variety, the actress said her co-star and friend wasn't struggling when he passed away.
"He was happy. He was healthy. He had quit smoking. He was getting in shape. He was happy — that’s all I know.
"I was literally texting with him that morning, funny Matty."
"I miss him dearly. We all do. Boy, he made us laugh really hard," she added.
